In short: Both are ways to define who sees an ad, but they draw on entirely different kinds of evidence. Custom Audiences are built from data you already own or activity people generated directly with your business - site visits, purchases, uploaded lists, video views - so membership is grounded in something that actually happened. Interest Targeting is built from Meta's inference about what a person generally engages with elsewhere on the platform, so it approximates affinity for people who have never interacted with you at all. Interest Targeting

The subset of Meta's detailed targeting built from topics and pages people engage with, used to approximate affinity when no first-party signal exists yet.

Interests are inferred from Facebook and Instagram activity, so they describe engagement patterns rather than verified intent, and Meta shows you nothing about how a user qualified. They earn their place at the top of the funnel when you have no pixel history to work from. The usual error is layering dozens of interests, producing an audience indistinguishable from broad targeting anyway.
