They power both retargeting and lookalike seeds. Custom Audiences require a data source to exist before you can use them; Detailed Targeting requires nothing but a category selection, which is why it is the only one of the two that works on a brand-new account.
You can inspect and defend exactly why someone is in a Custom Audience, since you defined the qualifying rule; you cannot do the same for Detailed Targeting, since Meta does not expose why a user matched an interest.
Custom Audiences decay through data mechanics - retention windows, stale uploads - while Detailed Targeting decays through catalog changes, since Meta periodically retires interest and behavior options without notice.
